Enrichment and Isolation Procedures

Thiorhodospira species can be selectively enriched under photo-autotrophic conditions with sulfide as the electron donor and in saline and alkaline media similar to those for Ectothiorhodospira and Halorhodospira species. From suitable environmental sources, isolation can be achieved using a basal medium1 in agar dilution series inoculated with natural samples without prior enrichment. Cultures are grown phototrophically in 50-ml screw-capped bottles at 25-30°C and a light intensity of 2000 lux.
