Enrichment and Isolation Procedures

Methods for enrichment and isolation are the same as those described for the genus Chromatium. Halochromatium species are generally found in microbial mats at the sediment surface of hypersaline environments, under salt (gypsum) deposits of salterns, in shallow salt lakes, and in evaporative lagoons with salinities of approx. 10-20%. From samples of purple layers in mats, these bacteria can be enriched or isolated by direct inoculation of deep agar dilution series in synthetic media containing 7-10% NaCl, as described by Caumette et al. (1988).
