Enrichment and Isolation Procedures

Thiococcus pfennigii is the only species of the family Chromatiaceae that contains bacteriochlorophyll b described so far. Owing to the presence of this bacteriochlorophyll, the bacterium has an in vivo absorption maximum in the infrared region of the spectrum, between 1020 nm and 1030 nm. By use of selective filters, this specific absorption has been successfully applied for the selective enrichment culture of Thiococcus species (Eimhjellen et al., 1967; Eimhjellen, 1970). Otherwise, the culture conditions and isolation procedures are the same as described for the genus Chromatium. The media described for Chromatium species can also be used for Thiococcus species, if supplemented with 1-3% NaCl as required.
